[目的]2,4-滴丁酸作为苯氧羧酸类除草剂在全球范围内被广泛使用。为研究2,4-滴丁酸与人血清白蛋白之间的相互作用,探究其对人体的毒性。[方法]选择人血清白蛋白作为考察对象,利用荧光光谱、圆二色光谱(CD)及傅里叶红外光谱(FT-IR)。[结果]1分子的2,4-滴丁酸能够与1分子的人血清白蛋白在人血清蛋白的site II(subdomain IIIA)部位上结合,形成复合物,从而导致蛋白构象发生改变。同时淬灭了蛋白的内源荧光,属于静态淬灭机制,此结合过程主要靠疏水作用力驱动。[结论]此研究有助于在分子层面上理解2,4-滴丁酸对人血清蛋白的作用机制,对于探讨2,4-滴丁酸在生物体内的累积性以及对人体潜在的影响有较为重要的意义。
[Objective] 2,4-D butyric acid is widely used as a phenoxycarboxylic acid herbicide in the world. In order to study the interaction between 2,4-D butyric acid and human serum albumin and to explore its toxicity to humans. [Method] Human serum albumin was selected as the object of study. Fluorescence, circular dichroism (CD) and Fourier transform infrared (FT-IR) spectra were used. [Result] One molecule of 2,4-D butyric acid could bind with one molecule of human serum albumin at the site of human serum protein II (subdomain IIIA) to form a complex, resulting in the conformational change of the protein. At the same time, the endogenous fluorescence of the protein is quenched, and belongs to the static quenching mechanism. The binding process is mainly driven by the hydrophobic force. [Conclusion] This study is helpful to understand the mechanism of action of 2,4-D butyric acid on human serum albumin at the molecular level, and to explore the potential accumulation of 2,4-D butyric acid in the body and its potential impact on human More important meaning.
