目的:以氧化低密度脂蛋白处理培养的兔主动脉平滑肌细胞(SMC)致细胞增殖,观察阿魏酸钠(SF)对SMC增殖的影响。方法:应用硫代巴比妥酸比色法测定脂质过氧化物(LPO)水平,氚胸腺嘧啶核苷(3HTdR)掺入实验观察SMC的DNA合成。结果:SF能明显降低培养液中LPO水平的升高,并呈剂量依赖性地减少SMC3HTdR的掺入量。结论:SF能明显抑制SMC增殖,其作用机制可能与抗脂质过氧化有关。
OBJECTIVE: To investigate the effect of sodium ferulate (SF) on the proliferation of SMCs in cultured rabbit aortic smooth muscle cells (SMC) treated with oxidized low-density lipoprotein. Methods: The level of lipid peroxidation (LPO) was determined by thiobarbituric acid colorimetry, and DNA synthesis of SMC was observed by incorporation of tritiated thymidine (3HTdR). RESULTS: SF could significantly reduce the increase of LPO in culture medium and reduce the incorporation of SMC3H-TdR in a dose-dependent manner. Conclusion: SF can significantly inhibit the proliferation of SMC, and its mechanism may be related to anti-lipid peroxidation.
