为探讨血管内皮生长因子 (VEGF)及其受体 (KDR)的表达与人大肠癌组织血管生成的关系 ,采用免疫组织化学SABC法观察了 6 8例人大肠癌组织中的VEGF及KDR的定位与分布 ,并对血管进行染色及计数。结果显示 ,6 8例大肠癌组织中VEGF表达阳性率为 5 5 9% (38/ 6 8) ,阳性物质阳性物质主要位于肿瘤细胞胞膜及胞浆 ;KDR表达阳性率为 4 5 6 % (31/ 6 8) ,既可位于癌组织及癌组织旁的血管内皮细胞 ,又可位于肿瘤细胞胞膜及胞浆。VEGF表达与大肠癌Dukes分期密切相关。VEGF表达阳性大肠癌组织的微血管密度(MVD)显著高于VEGF表达阴性者(P <0 0 1) ,而且随着VEGF表达强度的增强 ,癌组织内微血管密度明显增加 (P <0 0 1)。结果提示 ,大肠癌细胞分泌的VEGF既可以旁分泌的形式促进肿瘤血管的生成 ,也可能存在着自分泌形式 ;VEGF与大肠癌的生长、浸润和转移密切相关 ,是大肠癌主要的血管新生诱导因子之一 ,可促进大肠癌的血管生成
In order to investigate the relationship between the expression of vascular endothelial growth factor (VEGF) and its receptor (KDR) and angiogenesis in human colorectal cancer tissue, immunohistochemical SABC method was used to detect the expression of VEGF and KDR in 68 human colorectal cancer tissues And distribution, and blood vessels were stained and counted. The results showed that the positive rate of VEGF expression in 68 cases of colorectal cancer was 55.9% (38/68), positive substance was mainly located in the membrane and cytoplasm of tumor cells; the positive rate of KDR expression was 456% 31/68), both located in the cancer tissue and cancer tissue next to the vascular endothelial cells, but also located in the tumor cell membrane and cytoplasm. VEGF expression is closely related to Dukes stage of colorectal cancer. The microvessel density (MVD) of VEGF-positive colorectal cancer tissues was significantly higher than that of VEGF-negative ones (P <0.01), and the microvessel density was significantly increased with the increase of VEGF expression (P <0.01) . The results suggest that VEGF secreted by colorectal cancer cells may promote the formation of tumor blood vessels in the form of paracrine or autocrine forms.
