目的研究自噬相关蛋白Beclin-1在大鼠急性肾损伤伤后肾组织中的表达情况,并探讨其意义。方法选用24只雄性大鼠SD大鼠,体重250±30g,按随机数字表法分为两组,假手术组(Sham组)和缺血-再灌注组(I/R组);各组于再灌注后12、24、48h 3个相应时间点收获肾脏标本。HE染色观察肾脏病理变化,免疫组化法测定自噬相关蛋白Beclin-l的表达情况,透射电镜观察肾脏组织自噬现象。结果与假手术组比较,大鼠急性肾损伤48h,肾小管细胞Beclin-1蛋白的表达最强(P<0.05),再灌注损伤后48h,自噬体数量显著增多,细胞内出现染色质块状边集,核形态不规则的明显凋亡现象。结论大鼠急性肾损伤后自噬相关蛋白Beclin-1的表达上调,说明大鼠肾急性肾损伤后肾脏组组织自噬活性上调。
Objective To study the expression of autophagy-related protein Beclin-1 in renal tissues of rats with acute renal injury and its significance. Methods Twenty-four male Sprague-Dawley rats weighing 250 ± 30g were divided into two groups according to random number table: Sham group and ischemia / reperfusion group (I / R group) At 12, 24 and 48h after reperfusion, kidney samples were harvested at three corresponding time points. The pathological changes of kidney were observed by HE staining. The expression of autophagy-related protein Beclin-1 was detected by immunohistochemistry. Autophagy was observed by transmission electron microscope. Results Compared with the sham operation group, Beclin-1 protein expression was the highest (P <0.05) in renal tubular cells 48h after acute renal injury, and the number of autophagosomes was significantly increased 48h after reperfusion injury. Shaped edge set, the obvious irregular nuclear morphosis phenomenon. Conclusion The expression of autophagy-related protein Beclin-1 is up-regulated after acute kidney injury in rats, indicating that the renal autophagy activity is upregulated after acute kidney injury in rats.
