目的:探讨大黄附子成分配伍后对阳虚便秘大鼠的治疗作用及整合药效学特征。方法:采用山西白醋和0℃冰水活性炭建立大鼠阳虚便秘动物模型,随机分成模型空白组、附子模型组、配伍模型组;将正常大鼠分成正常空白组、附子正常组、配伍正常组,对模型空白组和正常空白组灌胃相应生理盐水,其他4组灌胃相应药物,采血。按照试剂盒说明书测定大鼠不同时间点血浆中胃动素(motillin,MTL)、胃泌素(gastrin,GT)、内皮素(endothelin,ET)、血管活性肠肽(vasoactive intestinal peptide,VIP)的浓度,并进行主成分分析。结果:配伍正常组与附子正常组比较,配伍模型组与附子模型组比较,大鼠各时间点血浆中MTL、GT、ET、VIP各自的浓度和四者的整合浓度均明显升高,且均在180 min时升高最明显,差异有统计学意义(P<0.05或P<0.01)。结论:附子总生物碱和大黄蒽醌配伍后可有效治疗大鼠阳虚便秘;MTL、GT、ET、VIP的整合药效学数据仍然具有明显的药效学特征,且整合后与整合前各成分的药效学特征基本一致,可表达上述4种成分整体的药效学特征。
Objective: To investigate the therapeutic effect and the integrated pharmacodynamic characteristics of rhubarb and aconite on constipated rats after Yang compatibility. Methods: The animal models of Yang constipation in rats were established by using Shanxi white vinegar and 0 ℃ ice water activated carbon and randomly divided into model blank group, aconite model group and compatibility model group. The normal rats were divided into normal blank group, aconite normal group and normal group , The model blank group and the normal blank group gavage the corresponding saline, the other four groups gavage the corresponding drug, blood collection. Conclusion: The combination of total alkaloids of Rhizoma Picrorhizae and Rhubarb anthraquinone can effectively treat constipation of yang deficiency in rats. The integrated pharmacodynamic data of MTL, GT, ET and VIP still have obvious pharmacodynamic characteristics. Pharmacodynamic characteristics of the ingredients are basically the same, can express the above four components of the overall pharmacodynamic characteristics.
