The nature of amyloidosis is not a sugar but a complex of a sugar-containing protein and a sulfated mucopolysaccharide. Electron microscopy is composed of two fibers (or fibrils). Clinically, Congo red stained tissue sections are usually used for observation, but the staining is different, and the contrast with the surrounding structures is not obvious, so it is not easy to confirm its presence or absence. If you look at the tissue section under a polarized light microscope, it is easy to distinguish. Positive substances (also known as starch-like substances) appear green or grass green on a dark background. In order to save the purchase price of polarized light microscopes, this article describes the preparation of polarizing plates and oscillating plates using polarizing films, using ordinary optical microscopes.
