目的:观察半夏水提取液对K562细胞增殖及凋亡的影响,初步探讨半夏对白血病的抗肿瘤作用机制。方法:采用CCK-8法,检测半夏水提取液不同浓度梯度范围内(35、350、700、1 000μg/ml)及不同作用时间(24、48、72h)对K562细胞的增殖抑制率;流式细胞仪检测K562细胞48h凋亡及周期变化。结果:半夏水提取液对人白血病K562细胞的增殖有明显的抑制作用,且呈剂量依赖性和时间依赖性;流式细胞仪分析结果显示半夏水提取液作用K562细胞48h后细胞凋亡百分率随半夏水提取液浓度增加而增加,且半夏水提取液可以诱导K562细胞周期阻滞在G0/G1期。结论:半夏水提取液在一定浓度范围内对人白血病细胞K562的增殖有抑制作用,且呈一定的量效时效关系;其对K562细胞的抑制作用可能与其诱导细胞凋亡和细胞周期阻滞有关。
OBJECTIVE: To observe the effect of aqueous extract of Pinellia ternata on the proliferation and apoptosis of K562 cells and to explore the anti-tumor mechanism of Banxia on leukemia. Methods: The inhibitory rate of proliferation of K562 cells was detected by CCK-8 assay in different concentration gradient (35,350,700,1 000μg / ml) and different time (24,48,72h) Flow cytometry analysis of K562 cells 48h apoptosis and cycle changes. Results: The extract of Pinellia ternata could inhibit the proliferation of human leukemia K562 cells in a dose-dependent and time-dependent manner. The results of flow cytometry showed that the apoptosis of K562 cells was inhibited The percentage increased with the increase of aqueous extract of Pinellia ternata, and the extract of Pinellia ternata could induce the cell cycle arrest of K562 in G0 / G1 phase. CONCLUSION: The extract of Pinellia ternata inhibits the proliferation of human leukemia cell K562 in a certain concentration range, and shows a dose-effect-time-dependence relationship. The inhibitory effect of the extract on K562 cells may be related to its induction of apoptosis and cell cycle arrest related.
