目的研究轮状病毒(Rotavirus,RV)P[8]G1和P[2]G3株灭活疫苗的免疫原性及不同血清型间的交叉识别作用。方法采用Vero细胞转瓶培养RV,经纯化灭活、佐剂吸附等工艺,制备RV P[8]G1和P[2]G3株灭活疫苗,分别经肌内免疫Wistar大鼠和猕猴,采用ELISA法和微量病毒中和试验检测大鼠和猕猴血清中RV特异性抗体效价和中和抗体效价,流式细胞术检测猕猴血液中CD4+和CD8+T细胞的数量。结果 P[8]G1和P[2]G3株疫苗样品的RV抗原含量分别为10 240和5 120 EU/ml;免疫2次后,P[8]G1和P[2]G3株疫苗诱导大鼠产生的血清IgG抗体GMT分别为(43 407.71±1 654.51)和(41 520.61±2 047.61),诱导猕猴产生的血清IgG抗体GMT分别为(13 654.45±3 125.27)和(10 350.68±997.83);单一血清型疫苗免疫后产生的中和抗体具有型间交叉识别作用;2株疫苗免疫猕猴后均能刺激其CD4+T淋巴细胞增殖,CD8+T淋巴细胞无明显变化。结论 2株RV灭活疫苗均具有良好的免疫原性及型间交叉识别作用。
Objective To study the immunogenicity and cross-recognition between different serotypes of rotavirus (RV) P [8] G1 and P [2] G3 inactivated vaccines. Methods RV RV [8] G1 and P [2] G3 inactivated vaccine were prepared by Vero cell culture in vitro and purified by inactivation and adjuvant adsorption. Wistar rats and macaques were immunized intramuscularly with The titer of RV-specific antibodies and the neutralizing antibody titer in the serum of rats and cynomolgus monkeys were detected by ELISA and micro-virus neutralization assay. The numbers of CD4 + and CD8 + T cells in blood of cynomolgus monkeys were detected by flow cytometry. Results The RV antigen contents of the vaccine strains P [8] G1 and P [2] G3 were 10 240 and 5 120 EU / ml, respectively. After 2 immunizations, the vaccine doses of P [8] G1 and P [2] The GMTs of serum IgG antibodies produced by mice were (43 407.71 ± 1 654.51) and (41 520.61 ± 2 047.61), respectively. The GMTs of serum IgG antibodies induced by rhesus monkeys were (13 654.45 ± 3 125.27) and (10 350.68 ± 997.83), respectively. The neutralizing antibodies produced by a single serotype vaccine had the cross-typed cross-recognition effect. Both of the two vaccines could stimulate the proliferation of CD4 + T lymphocytes and the CD8 + T lymphocytes did not change significantly. Conclusion Both RV inactivated vaccines have good immunogenicity and cross-recognition effect.
