目的:提供一种鉴别中药材海龙及其伪品的方法。方法:采用随机引物扩增DNA技术,对拟海龙、刁海龙、尖海龙、粗吻海龙、海蠋鱼、宝珈海龙进行基于Nei&Li’s相似系数的UPGMA聚类分析,并构建其鉴别模式。结果:从18条随机引物中筛选到LJ04,LJ09,LJ16,LJ19 4条引物能扩增出稳定可靠的条带。基于以上4条随机引物构建的聚类图能较好的从属水平区分各海龙样品,且与样品外观分析结果一致。基于LJ09,LJ19 2条引物的扩增结果构建的6种海龙的鉴别模式可以将海龙正品逐一鉴别出来。结论:随机引物扩增DNA技术能较好地鉴别海龙及其伪品。
Objective: To provide a method to identify the Chinese medicinal material Hailong and its counterfeit products. METHODS: The UPGMA cluster analysis based on Nei&Li’s similarity coefficient was carried out by using random primers to amplify DNA technology, and the identification models were constructed for Hydralonidae, Acrocephalosaurus, Acrocephalosaurus, Acrocephalus, Alocasia, Bream, and Bream. Results: Four primers of LJ04, LJ09, LJ16, and LJ19 were screened out from 18 random primers and a stable and reliable band was amplified. The cluster diagrams based on the above four random primers can better distinguish the sea dragon samples from the subordinate level and are consistent with the appearance analysis results of the samples. Based on the identification results of LJ09 and LJ19 2 primers, the identification patterns of 6 kinds of sea dragons can be identified one by one. Conclusion: Random primers amplified DNA technology can better identify sea dragons and their counterfeits.
