目的:探讨四磨汤有效成分槟榔碱、乌药醚内酯、橙皮苷、木香烃内酯对慢性应激小鼠血清Ghrelin、NO、Leptin含量的影响。方法:将144只小鼠随机分为正常组8只和造模组136只,模型小鼠给予不可预知的慢性应激造模21 d后随机分为模型组、四磨汤组及四磨汤成分组(15组)共17组;造模第15天起给药7 d后,酶联免疫吸附法测定血清NO、Ghrelin、Leptin含量。结果:模型组血清Ghrelin水平降低,NO、Leptin水平升高,与正常组及四磨汤组比较,差异均有统计学意义(P<0.01)。四磨汤有效成分3药及4药组合组Ghrelin水平升高,NO、Leptin水平降低,与模型组比较,差异均有统计学意义(P<0.05或P<0.01);而四磨汤有效成分单药组及两两组合组Ghrelin水平稍升高,NO、Leptin水平稍降低;与四磨汤组比较,差异有统计学意义(P<0.05或P<0.01);与模型组比较,差异均无统计学意义(P>0.05)。结论:槟榔碱、乌药醚内酯、橙皮苷、木香烃内酯可能为四磨汤的有效物质基础,四磨汤有效成分可以不同程度提高血清Ghrelin水平,降低血清NO、Leptin水平,可能是四磨汤调节胃肠功能的作用机制之一。
OBJECTIVE: To investigate the effects of Aretelmin, Wolteride, Hesperidin on the serum Ghrelin, NO and Leptin levels in chronic stress mice. Methods: 144 mice were randomly divided into normal group (n = 8) and model group (n = 136). The model mice were given unpredictable chronic stress model for 21 d and then randomly divided into model group, Si Mintang group (15 groups) were divided into 17 groups. Serum NO, Ghrelin and Leptin levels were determined by enzyme-linked immunosorbent assay after 7 days of administration on the 15th day after modeling. Results: The levels of Ghrelin, NO and Leptin in model group were significantly lower than those in normal group and Sihuangtang group (P <0.01). Sihurong active ingredients 3 and 4 drug combination group Ghrelin levels, NO, Leptin levels decreased, compared with the model group, the differences were statistically significant (P <0.05 or P <0.01); and Si mill soup active ingredients The level of Ghrelin increased slightly and NO and Leptin decreased slightly in the single drug group and the combination of two groups (P <0.05 or P <0.01), and the difference was statistically significant No statistical significance (P> 0.05). CONCLUSION: Arecoline, wolfberry ether lactone, hesperidin and woodlocolactone may be the effective material basis for Si mill soup. The active ingredients of Si mill decoction can increase serum Ghrelin level, decrease serum NO and Leptin level, May be the role of four mill soup regulate gastrointestinal function.
