目的:评价莱菔硫烷(SFN)与5-氟尿嘧啶(5-Fu)对人高转移唾液腺腺样囊性癌细胞(ACC-M)生长的抑制作用,同时检测参与诱导抗癌药物耐药性的核因子κB(NF-κB)的表达。方法:中位效应法评价SFN与5-Fu联用对ACC-M细胞增殖的影响,NF-κB p65表达采用Western印迹法检测。采用SPSS10.0软件包对所得数据进行t检验和方差分析。结果:SFN与5-Fu单独作用于ACC-M时的中位抑制浓度分别为(38.58±4.97)μmol/L和(120.90±12.96)μmol/L。两者联合使用时,对ACC-M细胞生长呈现协同抑制作用,并且引起细胞核NF-κB p65表达显著降低。与对照组相比,两药联合使用时,NF-κB p65的表达约降低5倍,P<0.05。结论:SFN与5-Fu联合使用可协同抑制ACC-M细胞增殖,该作用与SFN、5-Fu协同抑制细胞核NF-κB p65的表达有关。
Objective: To evaluate the inhibitory effect of sulforaphane (SFN) and 5-fluorouracil (5-Fu) on the growth of human highly metastatic salivary adenoid cystic carcinoma cells (ACC-M) Nuclear factor κB (NF-κB) expression. Methods: The median effect method was used to evaluate the effect of SFN combined with 5-FU on the proliferation of ACC-M cells. The expression of NF-κB p65 was detected by Western blotting. SPSS10.0 software package was used for t-test and ANOVA. Results: The median inhibitory concentrations of SFN and 5-Fu were (38.58 ± 4.97) μmol / L and (120.90 ± 12.96) μmol / L, respectively. When used in combination, they showed a synergistic inhibitory effect on the growth of ACC-M cells and markedly decreased the expression of NF-κB p65 in the nucleus. Compared with the control group, the combination of the two drugs, NF-κB p65 expression decreased about 5 times, P <0.05. Conclusion: The combination of SFN and 5-Fu can synergistically inhibit the proliferation of ACC-M cells, which is related to the synergistic effect of SFN and 5-Fu on the expression of NF-κB p65.
