目的观察尾吊模拟失重对大鼠颈总动脉和股动脉自噬活性的影响。方法雄性SD大鼠,随机分为尾吊组和对照组,通过Western Blot、PCR及免疫组化检测尾吊模拟失重后大鼠颈总动脉及股动脉微管相关蛋白轻链3(LC3)和Bcl-2相关蛋白(Beclin-1)的表达变化。结果尾吊28 d后,大鼠股动脉LC3及Beclin-1的mRNA和蛋白的表达均显著高于对照组,免疫组化发现LC3在内皮层及平滑肌层均有表达升高;而颈总动脉的LC3及Beclin-1的mRNA和蛋白的表达无明显变化。结论尾吊模拟失重可以诱导大鼠股动脉自噬活性增强。
Objective To observe the effect of tail-suspended weightless-loading on the autophagic activity of common carotid artery and femoral artery in rats. Methods Male Sprague-Dawley rats were randomly divided into tail suspension group and control group. Western blot, PCR and immunohistochemistry were used to detect the expression of LC3 in the common carotid artery and femoral artery of rats after simulated weightlessness Bcl-2-related protein (Beclin-1) expression changes. Results After 28 days, the mRNA and protein expressions of LC3 and Beclin-1 in femoral artery of rats were significantly higher than those in control group. The expression of LC3 in both endothelial and smooth muscle layers was found by immunohistochemistry. The common carotid arteries LC3 and Beclin-1 mRNA and protein expression no significant change. CONCLUSION: Simulated weightlessness in the tail suspension can induce autophagy in the femoral artery of rats.
