目的采用基于核磁共振的代谢组学方法研究高果糖喂养对白脂肪代谢组的影响。方法选取16只Wistar大鼠,将其适应1周后随机分为正常对照组和模型组,每组8只。采用100 g·L-1果糖水饲喂8周建立胰岛素抵抗(insulin resistance,IR)模型,对照组给予与模型组等体积的纯净水。8周末取大鼠白脂肪组织,运用主成分方法(principal component analysis,PCA)对白脂肪组织水溶性提取液的核磁共振氢谱(1H-NMR)谱进行分析。结果与正常对照组比较,模型组大鼠的白脂肪组织代谢组明显变化,其中丙氨酸、氧化三甲胺/甜菜碱、胆碱和丙三醇含量升高,乳酸、脂质、谷氨酸、甘油磷脂胆碱/磷脂胆碱和不饱和脂类含量降低,差异均有统计学意义(P<0.05,P<0.01)。结论采用核磁共振代谢组学方法研究高果糖对白脂肪代谢组的影响,阐明了IR状态下白脂肪组织提取液的代谢特征及其相应生化代谢过程的改变。
OBJECTIVE: To study the effect of high fructose diet on white fat metabolism by nuclear magnetic resonance based metabonomics. Methods Sixteen Wistar rats were selected and randomly divided into normal control group and model group after one week of acclimation, with 8 rats in each group. Insulin resistance (IR) model was established by feeding with 100 g · L-1 fructose water for 8 weeks. The control group was given equal volume of pure water to the model group. At the end of the 8th week, white adipose tissue was obtained from rats, and the 1H-NMR spectra of water-soluble extract of white adipose tissue were analyzed by principal component analysis (PCA). Results Compared with the normal control group, the metabolism of the white adipose tissue in the model group was significantly changed, in which the contents of alanine, trimethylamine oxide / betaine, choline and glycerol were elevated, and the content of lactate, lipid, glutamic acid , The content of glycerophospholipid choline / phospholipid choline and unsaturated lipids decreased, the differences were statistically significant (P <0.05, P <0.01). Conclusion The effect of high fructose on white fat metabolism was studied by using nuclear magnetic resonance metabonomics method. The metabolic characteristics of white fat tissue extract and its corresponding biochemical metabolic process under IR were clarified.
