目的探讨亲水性琼脂糖包埋的大鼠乳腺癌细胞生长上清对人乳腺癌细胞生长的影响。方法应用亲水性琼脂糖包埋大鼠乳腺癌SHZ-88细胞,使其在凝胶内部生长。通过中性红染色显微镜下观察凝胶内生长细胞的活性。收集不同培养时间的上清,作用于人乳腺癌MCF-7细胞,通过CCK-8检测作用不同时间后的细胞活存状况。结果通过中性红染色后显微镜观察,在包埋SHZ-88细胞的凝胶内可见散在的死细胞与成团的活细胞。包埋细胞生长的上清能够抑制人MCF-7细胞生长,包埋细胞生长时间越长获得的上清抑制细胞生长能力越强。结论亲水性琼脂糖包埋的大鼠乳腺癌细胞生长上清能够有效抑制人MCF-7细胞生长。
Objective To investigate the effect of hydrophilic agarose-embedded rat breast cancer cell supernatant on the growth of human breast cancer cells. Methods SHZ-88 breast cancer cells were embedded in hydrophilic agarose to grow inside the gel. The growth of cells in the gel was observed under a neutral red staining microscope. The supernatants of different culture time were collected to act on human breast cancer MCF-7 cells, and the cell viability after different time was tested by CCK-8. The results were observed by neutral red staining microscopy, embedded in the SHZ-88 cells embedded in the gel scattered loose dead cells and agglomeration of living cells. The supernatant of the embedded cells can inhibit the growth of human MCF-7 cells. The longer the time of the embedded cells, the higher the supernatant can inhibit the cell growth. Conclusion Hydrophilic agarose-embedded rat breast cancer cell supernatant can effectively inhibit the growth of human MCF-7 cells.
