目的观察磷酸西格列汀对2型糖尿病(T2DM)大鼠肝脏组织亲吻素1(KISS1)水平的影响。方法 20只大鼠高脂饮食加小剂量链脲佐菌素诱导构建T2DM大鼠模型后均分为两组:实验(SIT)组实施磷酸西格列汀10mg·kg~(-1)·d~(-1)灌胃,模型(DM)组给予等体积生理盐水灌胃。另取10只大鼠(NC组)常规喂养,等体积生理盐水灌胃。干预12周后,采用qRT-PCR和Western blot分别检测各组大鼠肝组织KISS1 mRNA水平和蛋白表达。结果与NC组相比,DM组大鼠KISS1 mRNA和蛋白水平提高(P<0.05)。SIT组大鼠KISS1 mRNA和蛋白水平低于DM组(P<0.05)。结论磷酸西格列汀可有效降低T2DM大鼠肝脏组织KISS1水平的升高。
Objective To observe the effect of sitagliptin phosphate on the level of KISS1 in liver of type 2 diabetic rats (T2DM). Methods Twenty rats with high-fat diet plus low-dose streptozotocin (STZ) -induced T2DM rats were divided into two groups: experimental (SIT) group received sitagliptin phosphate 10 mg · kg -1 ~ (-1) gavage, and the model group (DM) was given equal volume of normal saline. Another 10 rats (NC group) routine feeding, equal volume of normal saline gavage. After intervention for 12 weeks, the expression of KISS1 mRNA and protein in liver tissue of each group were detected by qRT-PCR and Western blot respectively. Results Compared with NC group, the level of KISS1 mRNA and protein in DM group increased (P <0.05). The level of KISS1 mRNA and protein in SIT group was lower than that in DM group (P <0.05). Conclusion Sitagliptin phosphate can effectively reduce the level of KISS1 in liver of T2DM rats.
