目的探讨珍宝丸预处理对缺氧性脑损伤新生乳鼠大脑白质髓鞘碱性蛋白(MBP)表达的影响。方法取新生1 d Wistar大鼠210只,雌雄不限,随机分为对照组、模型组和珍宝丸组各70只,每组又分为0.5、24、48、72 h及5、7和14 d 7个时间点各10只。新生1 d的Wistar大鼠均于第1天开始灌胃持续至实验结束,对照组和模型组灌胃给予生理盐水,珍宝丸组灌胃给予珍宝丸;第7天将乳鼠置于8%O2+92%N2(V/V)的缺氧箱内2 h建立新生鼠脑缺氧模型,分别于缺氧后0.5、24、48、72 h及5、7和14 d处死取脑组织待测。采用HE染色检测乳鼠脑组织白质的病理改变,采用ELISA法检测各组乳鼠血清中MBP的蛋白含量,采用免疫组织化学方法检测各组乳鼠脑白质中MBP蛋白的阳性表达。结果对照组MBP的表达随着时间点0.5、24、48、72 h阳性表达逐渐增强(P<0.05),72 h后各时间点差异无统计学意义(P>0.05);与对照组比较,模型组和珍宝丸组MBP的表达随着时间点0.5、24、48、72 h明显阳性表达增强,模型组高于珍宝丸组且均在72 h时间点达高峰,差异有统计学意义(P<0.05),5、7、14 d随着时间点的延续逐渐降低,至14 d与对照组差异无统计学意义(P>0.05)。结论珍宝丸的预处理可通过早期下调乳鼠MBP的蛋白水平,缓解各时间点缺氧对乳鼠脑白质的损伤,从而对缺氧性脑损伤具有一定的保护作用。
Objective To investigate the effects of Zhenbao Pills preconditioning on the expression of MBP in neonatal neonatal rats with hypoxic brain damage. Methods Two hundred and twenty fresh Wistar rats were randomly divided into control group, model group and Zhenbao pill group, with each group being divided into two groups: 0.5, 24, 48, 72 h and 5, 7 and 14 d 7 time points of the 10. The expression of MBP protein in the neonatal rats was detected by immunohistochemistry. Results The expression of MBP in control group increased gradually at 0.5,24,48,72 h (P <0.05), and there was no significant difference at each time point after 72 h (P> 0.05). Compared with control group, The expression of MBP in model group and Zhenbao pill group increased obviously at 0.5,24,48 and 72 hrs, the model group was higher than that of Zhenbao pill group, and both reached the peak at 72 h, the difference was statistically significant (P <0.05). At 5, 14 days and 14th days, the expression of MMP-9 decreased with the time point, and there was no significant difference between the 14th and the control group (P> 0.05). Conclusion Zhenbao pill pretreatment can reduce the level of MBP protein in neonatal rats and alleviate the damage of white matter of neonatal rats at different time points, which can protect hypoxic brain injury.
