目的动态观察单侧输尿管梗阻大鼠(UUO)模型肾间质炎性损害病理改变及核因子-κB(NF-κB)、转化生长因子-β1(TGF-β1)在肾间质的表达,探讨血必净对肾间质炎性损害的干预机制。方法Sprague-Dawley(简称SD)清洁级健康大鼠雌雄各半共60只,随机分为三组:假手术组(SOR组)、模型组(MOD组)、血必净治疗组(XBJ组),每组20只;建立UUO模型,分别于手术后第7d、第14d各组随机选取10只大鼠,取梗阻侧肾组织,行HE、MASSON染色及免疫组化染色。观察肾脏间质病理变化以及NF-κB p65和TGF-β1在肾间质的表达。结果血必净能减轻UUO大鼠肾间质炎性损害(第7d、第14d肾小管间质损伤指数、肾间质胶原面积P<0.05);血必净能下调肾组织间质NF-κBp65及TGF-β1的的表达(P<0.05)有统计学意义。结论血必净减轻肾间质炎性损害,其机制可能通过抑制肾组织间质NF-κB p65及TGF-β1的的表达,并对早期肾间质纤维化有抑制作用。
Objective To observe the pathological changes of renal interstitial inflammatory lesions and the expression of nuclear factor-κB (NF-κB) and transforming growth factor-β1 (TGF-β1) in renal interstitium in rats with unilateral ureteral obstruction (UUO). Xue Bi Jing Intervention Mechanism of Renal Interstitial Inflammatory Damage. Methods Sixty Sprague-Dawley (abbreviated as SD) clean grade healthy rats were divided into three groups randomly: sham operation group (SOR group), model group (MOD group), Xuebijing treatment group (XBJ group). Twenty rats in each group were established. UUO models were established. Ten rats were randomly selected from each group on the 7th and 14th day after operation. Obstructed lateral kidney tissues were taken and HE, MASSON staining and immunohistochemical staining were performed. The pathological changes of renal interstitium and the expression of NF-κB p65 and TGF-β1 in renal interstitium were observed. Results Xuebijing could reduce the renal interstitial inflammatory lesions in UUO rats (renal tubular interstitial injury index, renal interstitial collagen area at the 7th and 14th day P<0.05); Xuebijingeng could down-regulate renal interstitial NF-κB p65 And the expression of TGF-β1 (P<0.05) was statistically significant. Conclusion Xuebijing can reduce renal interstitial inflammatory lesions. The mechanism may be through inhibiting the expression of NF-κB p65 and TGF-β1 in renal interstitial and inhibiting early renal interstitial fibrosis.
