目的探讨哺乳期染毒双酚A对子代雄鼠成年后睾丸结构及雄激素受体(AR)表达影响。方法 20只妊娠母鼠,待分娩后随机分为双酚A高、中、低剂量组(5、0.1、0.01 mg/kg)和对照组。母鼠分娩后第3 d开始每天给小鼠颈部皮下注射相应剂量的双酚A和玉米油,持续到第21 d断奶,在第70 d处死并取材,记录体重及睾丸重量,制作精子涂片,计算精子畸形率,观察小鼠睾丸组织形态学变化,采用免疫组织化学和Western blot法检测睾丸内AR表达。结果双酚A高剂量组成年雄鼠睾丸指数(0.44±0.018)明显低于对照组(0.64±0.039)(P<0.05);双酚A低、中、高剂量组成年雄鼠精子畸形率分别为(13.34±0.72)%、(16.41±0.67)%、(17.55±0.61)%,高于对照组的(2.07±0.24)%(P<0.05),呈剂量效应关系;双酚A组雄鼠睾丸曲细精管直径与生精上皮厚度均明显减小(P<0.05),各级生精细胞形态无明显改变;双酚A低、中、高剂量组雄鼠AR表达分别为(0.6642±0.0141)、(0.6363±0.0062)、(0.2938±0.2522),高于对照组(0.2123±0.2793)(P<0.05)。结论哺乳期接触一定剂量双酚A对雄鼠睾丸发育具有明显影响,精子畸形率增加,曲细精管直径减小,生精上皮变薄,睾丸内AR表达增加。
Objective To investigate the effect of lactational exposure to bisphenol A on the structure of testis and the expression of androgen receptor (AR) in adult offspring. Methods Twenty pregnant pregnant rats were randomly divided into high, medium and low doses of bisphenol A (5,0.1,0.01 mg / kg) and control group. Mice were injected subcutaneously with the corresponding dose of bisphenol A and corn oil subcutaneously on the neck of the mice on the 3rd day after childbirth. The animals were weaned until the 21st day and were sacrificed on the 70th day. The body weight and testicular weight were recorded to make sperm The sperm deformity rate was calculated. Morphological changes of testis were observed. The expression of AR in testis was detected by immunohistochemistry and Western blot. Results The testis index (0.44 ± 0.018) in male male rats with high dose of bisphenol A was significantly lower than that of the control group (0.64 ± 0.039) (P <0.05). The sperm deformity rates of male, male, male, (13.34 ± 0.72)%, (16.41 ± 0.67)% and (17.55 ± 0.61)% respectively, which was higher than that of the control group (2.07 ± 0.24)% (P <0.05) The diameter of seminiferous tubules and the thickness of seminiferous epithelium were significantly decreased (P <0.05), and the morphology of spermatogenic cells at all levels did not change significantly. The expression of AR in low, middle and high dose of bisphenol A group was (0.6642 ± 0.0141), (0.6363 ± 0.0062), (0.2938 ± 0.2522), higher than the control group (0.2123 ± 0.2793) (P <0.05). Conclusions Exposure to a dose of bisphenol A during lactation has a significant effect on testicular development in male rats. The sperm deformity rate increases, the diameter of seminiferous tubules decreases, the seminiferous epithelium becomes thinner, and the expression of AR in testis increases.
