目的研究硝苯吡啶(nihedipinec,Nif)和维拉帕米(verapamil,Ver)对人胚肺成纤维细胞(humanlungfibroblasts,HLF)的DNA和胶原合成的影响,从细胞水平评价钙拮抗剂防治器官纤维化并提供实验依据。方法采用MTT比色法、3H-TdR和3H-脯氨酸掺入法测定细胞增殖、DNA合成和胶原合成。结果Nif和Ver在0~50μmol/L浓度范围内均以浓度依赖方式抑制HLF增殖、DNA合成及胶原合成(P<0.05~0.01)。结论Nif和Ver具有抗纤维化作用
Objective To investigate the effects of nifedipine and verapamil on DNA and collagen synthesis in human lung fibroblasts (HLF). To evaluate the effect of calcium antagonists on the prevention and treatment of organ fibrosis And provide experimental basis. Methods Cell proliferation, DNA synthesis and collagen synthesis were determined by MTT assay, 3H-TdR and 3H-proline incorporation. Results Nif and Ver inhibited HLF proliferation, DNA synthesis and collagen synthesis (P <0.05 ~ 0.01) in a concentration-dependent manner in the concentration range of 0-50 μmol / L. Conclusion Nif and Ver have anti-fibrosis effects
