目的研究人直肠癌和结肠癌的肿瘤浸润 γδT淋巴细胞 ( γδTIL s)体外抗肿瘤活性。方法用固相抗体法体外扩增获得γδTIL s,并用 MTT法和 3H- Td R掺入实验体外观察γδTIL s对同种异体和异种肿瘤细胞的细胞毒及增殖反应。结果所获得γδTIL s对所测定的肿瘤细胞显示出明显的细胞毒活性。该杀伤效应可为 IL - 2 ,IL - 4,IL - 12 ,IL - 15 ,TNF-α和 IFN -γ所增强 ,而不受 IL - 10 ,GM- CSF和 TGF-β所影响。某些去增殖活性的肿瘤细胞可诱导静息化的γδTIL s产生显著的增殖反应。结论 γδTIL s可通过其强烈的肿瘤细胞毒参与机体的抗肿瘤免疫应答
Objective To study the in vitro antitumor activity of tumor infiltrating γδT lymphocytes (γδTIL s) in human colorectal cancer and colon cancer. Methods γδTIL s were obtained by in vitro amplification using solid-phase antibody method. Cytotoxicity and proliferative responses of γδTILs to allogeneic and xenogeneic tumor cells were observed by MTT assay and 3H-Td R incorporation experiments. As a result, the γδTILs obtained exhibited significant cytotoxic activity against the measured tumor cells. The killing effect can be enhanced by IL-2, IL-4, IL-12, IL-15, TNF-α and IFN-γ, but not by IL-10, GM-CSF and TGF-β. Certain deproliferating tumor cells can induce a proliferative response to resting γδTILs. Conclusion γδTIL s can participate in the anti-tumor immune response of the body through its strong tumor cytotoxicity
