目的:探讨大孔吸附树脂对女贞子中齐墩果酸吸附纯化的初步研究。方法:采用大孔树脂法分离纯化女贞子中齐墩果酸,并应用高效液相色谱法,以齐墩果酸为考核指标,筛选大孔吸附树脂类型,并对洗脱溶剂的选择、树脂柱径高比、洗脱流速、药材与树脂用量比、洗脱剂用量等进行考察。结果:以HPD-600为最优型号树脂用于分离纯化女贞子中齐墩果酸,以90%乙醇,树脂柱径高比为1:3,洗脱流速4BV·h-1,药材树脂用量比1:7为最佳工艺。结论:采用大孔树脂法分离纯化女贞子中齐墩果酸,并应用高效液相色谱法测定齐墩果酸含量,证明该法具有简便、快速、准确等优点。
Objective: To investigate the adsorption and purification of oleanolic acid from Ligustrum lucidum by macroporous resin. METHODS: Macroporous resin was used to separate and purify oleanolic acid from Ligustrum lucidum. HPLC method was used to evaluate the type of macroporous resin and the choice of elution solvent. The diameter ratio of the resin column, the elution flow rate, the ratio of the amount of the medicinal material to the resin, and the amount of the eluent were examined. Results: HPD-600 was the best resin for separating and purifying oleanolic acid in Ligustrum lucidum, with 90% ethanol, resin column diameter ratio of 1:3, elution flow rate 4BV·h-1, medicinal resin The ratio of 1:7 is the best process. Conclusion: The method of separation and purification of oleanolic acid from Ligustrum lucidum by macroporous resin method and the determination of oleanolic acid by high performance liquid chromatography show that the method is simple, rapid and accurate.
