目的 观察褪黑素对人肝癌 SMMC- 772 1细胞生长抑制作用及其作用机理。方法 采用不同浓度的褪黑素通过体外细胞培养、MTT显色技术研究其抑制作用 ,以流式细胞仪和电子显微镜技术观察其作用机理。结果 褪黑素对人肝癌 SMMC- 772 1细胞具有抑制作用 ,其 IC30 、IC50 分别为 1 .1 0和 2 .0 0 mmol/L。以 IC30 的褪黑素处理 SMMC- 772 1细胞 ,使其倍增时间由 2 1 .1 h延长到 38.4h。流式细胞仪观察到褪黑素导致 SMMC- 772 1细胞 G0 /G1期比例升高 ,S期比例降低。透射电镜可观察到以 IC30 的褪黑素处理的细胞 ,其超微结构发生了退行性改变。结论褪黑素对 SMMC- 772 1细胞具有抑制作用 ,其作用机理可能是褪黑素导致细胞 G0 /G1阶段的推迟进行
Objective To observe the inhibitory effect of melatonin on human hepatocellular carcinoma SMMC-772 1 cells and its mechanism of action. Methods The effects of different concentrations of melatonin were studied by in vitro cell culture and MTT coloration techniques. The mechanism of action was observed by flow cytometry and electron microscopy. Results Melatonin inhibited human hepatoma SMMC-772 1 cells with an IC30 and IC50 of 1.1 and 2.0 mmol/L, respectively. Treatment of SMMC-7721 cells with IC30 melatonin increased the doubling time from 21.1 h to 38.4 h. Flow cytometry observed that melatonin caused an increase in the ratio of G0/G1 phase in SMMC-7721 cells and a decrease in the proportion of S phase. Transmission electron microscopy showed that the melanin-treated cells of IC30 had degenerative changes in their ultrastructure. Conclusion Melatonin has an inhibitory effect on SMMC-7721 cells, and the mechanism may be that melatonin causes a delay in the G0/G1 phase of cells.
