目的:优化美花石斛多糖的提取工艺并对其含量进行测定。方法:采用紫外-可见分光光度法测定美花石斛多糖含量,以D-无水葡萄糖标准品为对照,检测波长为489nm。通过单因素考察及正交试验确定美花石斛多糖的最佳提取工艺并测定其含量。结果:采用水加热回流法对美花石斛多糖进行提取,确定最佳提取工艺条件为:料液比1∶80,提取时间1.5h,提取次数为2次;在此条件下测得不同产地美花石斛多糖的含量在6.18%~10.48%之间。结论:该方法结果准确、可靠,可作为美花石斛多糖的提取方法,为其质量控制提供了依据。
Objective: To optimize the extraction process of polysaccharides from Dendrobium chrysanthum and determine its content. Methods: The polysaccharide content of Dendrobium chrysanthum was determined by UV - visible spectrophotometry. The D - anhydrous glucose standard was used as the control. The detection wavelength was 489 nm. Through single factor investigation and orthogonal experiment, the best extraction technology of Dendrobium chrysanthum polysaccharide was determined and its content was determined. Results: The extraction of polysaccharides from Dendrobium chrysanthum was carried out by water heating and refluxing method. The optimal extraction conditions were determined as follows: the ratio of solid to liquid was 1:80, the extraction time was 1.5h and the number of extraction was 2 times. Under these conditions, Dendrobium polysaccharide content of 6.18% ~ 10.48%. Conclusion: The method is accurate and reliable, which can be used as the extraction method of polysaccharides from Dendrobium chrysanthum and provide the basis for its quality control.
