为鉴定长春市某渔场鲫鱼病原菌,从患病鲫鱼脾中分离到病原菌J YX-1,通过生理生化试验,16S rRNA、管家基因及毒力基因扩增,系统发育分析和药敏试验进行鉴定。结果显示,分离菌J YX-1对鲫鱼具有致病性;基于16Sr RNA与4种管家基因构建的系统发育树显示,该菌与维氏气单胞菌同属一个分支,亲缘关系较近,表明J YX-1为维氏气单胞菌;毒力基因检测发现,能从J YX-1扩增出气溶素(aer)、细胞毒性肠毒素(act)、黏附素(Aha)、丝氨酸蛋白酶(ser)、核酸酶(exu)及脂肪酶(lip)6种毒力因子基因。药敏试验结果显示,分离菌J YX-1对克拉霉素、妥布霉素及链霉素等11种药物敏感。本研究结果为鲫鱼感染维氏气单胞菌的鉴定及治疗提供了理论依据。
To identify pathogenic bacteria of crucian carp in a fishing ground of Changchun City, the pathogen J YX-1 was isolated from diseased crucian carp spleen and identified by physiological and biochemical tests, 16S rRNA, housekeeping gene and virulence gene amplification, phylogenetic analysis and susceptibility testing. The results showed that the isolate JYX-1 was pathogenic to crucian carp. Phylogenetic tree based on 16Sr RNA and four housekeeping genes showed that the strain belonged to the same branch of Aeromonas sobria and had a close genetic relationship, indicating that J YX-1 is Aeromonas sobria; virulence gene test found that it can amplify aer, cytotoxic enterokinase (Aha), serine protease ser), nuclease (exu) and lip (lip) six virulence genes. Susceptibility test results showed that isolates J YX-1 sensitive to 11 drugs such as clarithromycin, tobramycin and streptomycin. The results of this study provide a theoretical basis for identification and treatment of Aeromonas sobria infection in crucian carp.
