目的　探讨不同浓度高渗盐液对左心室舒张和收缩功能的影响。方法　 (1)用不同钠离子浓度的灌流液对离体大鼠心脏进行顺性灌注 2 0分钟 ,随后用等渗灌流液灌注 5分钟 ,连续观察心排量及左心室功能的变化 ,并测心肌含钙量 ;(2 )采用膜片钳技术观察 2 5 5mmol/LNaCl台氏液对豚鼠单一心肌细胞膜钙电流的作用。结果　 (1)含 16 5mmol/LNa+ 的灌流液可明显增加心排量 ,改善心脏的收缩舒张功能 ;而用高于 2 15mmol/LNa+ 的灌流液灌流时最初有短暂却十分明显的心脏收缩舒张功能障碍 ,但 5～ 10分钟后恢复到正常水平以上 ;各组在高渗灌流液换为等渗液后心脏收缩舒张功能都非常显著地增强。 (2 )高渗钠溶液明显增强豚鼠心肌细胞膜钙离子电流。结论　高渗盐液能直接作用于心脏 ,增强心肌细胞膜的钙内流 ,在一定的条件下可明显改善左心室收缩舒张功能 Objective To investigate the effects of different concentrations of hypertonic saline on left ventricular diastolic and systolic function. Methods (1) Peripheral perfusion of isolated rat hearts were perfusion with different concentrations of sodium ions for 20 minutes followed by perfusion with isotonic perfusate for 5 minutes. The changes of cardiac output and left ventricular function were observed continuously and measured (2) Patch-clamp technique was used to observe the effect of 25 5 mmol / L Taq solution on the single cardiomyocyte calcium currents in guinea pigs. Results (1) The perfusate containing 16 5mmol / L Na + could significantly increase the cardiac output and improve the systolic and diastolic function of the heart. When perfusion with more than 215mmol / L Na + was initially perforated, the transient but significant systolic and diastolic function However, it returned to normal levels after 5 to 10 minutes. The systolic and diastolic functions of all the groups were significantly enhanced after the hyperosmolar perfusion fluid was changed to isotonic solution. (2) Hypertonic sodium solution significantly increased the calcium current in guinea pig cardiomyocytes. CONCLUSION: Hypertonic saline can directly act on the heart and enhance the calcium influx in myocardial cell membrane. Under certain conditions, systolic and diastolic function of left ventricle can be obviously improved
