目的通过检测口腔鳞状细胞癌(OSCC)组织中全长型脾酪氨酸激酶[SYK(L)]的表达、与淋巴结转移的相关性分析,探讨SYK(L)对OSCC发生发展和转移的影响。方法采用实时荧光定量聚合酶链反应(RT-qPCR)、免疫印迹(Western blot)、免疫组织化学法检测SYK(L)在27例OSCC及其癌旁组织中表达的差异,并采用14例正常口腔黏膜作为对照,探讨其与OSCC发生及转移的关系。结果 OSCC组织中SYK(L)表达量明显低于正常牙龈组织(P<0.01),同一患者癌组织中SYK(L)表达量比癌旁组织低(P<0.05);淋巴结转移组患者的SYK(L)表达量普遍低于未转移组患者(P<0.05)。结论 SYK(L)可能与OSCC的发生和发展有关,作为抑癌基因在肿瘤的淋巴结转移过程中起至关重要的作用。
Objective To investigate the correlation between the expression of SYK (L) and the lymph node metastasis in oral squamous cell carcinoma (OSCC) and to explore the role of SYK (L) in the development and metastasis of OSCC influences. Methods The expression of SYK (L) in 27 cases of OSCC and adjacent normal tissues was detected by RT-qPCR, Western blot and immunohistochemistry. 14 cases of normal Oral mucosa as a control to explore the relationship with the occurrence and metastasis of OSCC. Results The expression of SYK (L) in OSCC tissues was significantly lower than that in normal gingival tissues (P <0.01). The expression of SYK (L) in the same patients was lower than that in adjacent tissues (P <0.05) (L) expression levels were generally lower than those without metastasis (P <0.05). Conclusion SYK (L) may be related to the occurrence and development of OSCC, and plays an important role as a tumor suppressor gene in the process of lymph node metastasis of tumors.
