目的检测APRG1基因mRNA在乳腺癌,癌旁组织及良性肿瘤周围正常组织中的表达,探讨APRG1基因表达与乳腺癌及其病程进展之间的关系。方法应用RT-PCR检测78例乳腺癌组织,癌旁组织及32例正常组织标本中APRG1基因mRNA表达。结果APRG1mRNA在恶性肿瘤组织中的表达较低。随TNM分期增加而下降,TNMⅢ期病人与Ⅰ期相比有统计学意义(P<0.05)。APRG1mRNA表达在癌细胞核分级3级的肿瘤标本较低,与1级相比有统计学意义(P<0.05)。结论APRG1基因的表达在乳腺癌组织、尤其病程较晚的肿瘤组织中降低,提示APRG1是一个抑癌基因。
Objective To detect the expression of APRG1 mRNA in breast cancer tissues, pericarcinoma tissues and normal tissues around benign tumors, and to explore the relationship between APRG1 gene expression and the progression of breast cancer. Methods RT-PCR was used to detect the mRNA expression of APRG1 in 78 cases of breast cancer tissues, adjacent non-cancerous tissues and 32 normal tissues. Results The expression of APRG1 mRNA in malignant tumors was low. With the TNM staging increased and decreased, TNM Ⅲ patients compared with the first phase was statistically significant (P <0.05). The expression of APRG1mRNA in tumor grade 3 was lower, which was statistically significant compared with grade 1 (P <0.05). Conclusion The expression of APRG1 gene is down-regulated in breast cancer tissues, especially in later-stage tumors, suggesting that APRG1 is a tumor suppressor gene.
