目的:探讨美他多辛对雄性酒精性脂肪肝大鼠肝组织中CYP2E1表达的影响。方法:30只清洁级Wistar大鼠随机分为3组:脂肪肝组梯度给予不同浓度的酒精灌胃8周;预防组于酒精灌胃同期给予美他多辛预防;对照组给予普通饮食。8周酒精灌胃结束后,B超检测肝组织影像学变化、病理学观察酒精性脂肪肝的形成以及免疫组织化学检测肝组织中CYP2E1的表达。结果:雄性酒精性脂肪肝大鼠模型建立成功;脂肪肝组、预防组与对照组比,CYP2E1表达均上调,P<0.05;脂肪肝组与预防组比,CYP2E1表达上调,P<0.05。结论:CYP2E1表达的上调参与了酒精性脂肪肝的发病过程,美他多辛可有效预防酒精性脂肪肝的发生及CYP2E1表达的上调。
Objective: To investigate the effect of metaodoxin on the expression of CYP2E1 in the liver of male alcoholic fatty liver rats. Methods: Thirty Wistar rats of clean grade were randomly divided into three groups: the rats in fatty liver group were given different concentrations of alcohol for 8 weeks; the prophylaxis group were given metadoxine prophylaxis in the same period of alcohol administration; the control group were given normal diet. After 8 weeks of alcohol instillation, B-mode ultrasonography was used to detect the changes of liver tissue. The pathological changes of alcohol-induced fatty liver were observed, and the expression of CYP2E1 in liver tissue was detected by immunohistochemistry. Results: Compared with control group, the expression of CYP2E1 in fatty liver group was significantly higher than that in control group (P <0.05). The expression of CYP2E1 in fatty liver group was higher than that in prevention group (P <0.05). CONCLUSION: Upregulation of CYP2E1 expression is involved in the pathogenesis of alcoholic fatty liver. Metatoxin can effectively prevent the occurrence of alcoholic fatty liver and up-regulate the expression of CYP2E1.
