本研究以还原型辅酶Ⅱ黄递酶组织化学法对大鼠胆道感染体内模型中肝细胞表达诱生型一氧化氮合酶(induciblenitricoxidesynthase,iNOS)的情况进行了研究,以了解这种条件下肝细胞是否有iNOS的表达及其表达的规律。方法:Wistar大鼠30只,随机分为5组,对照1组,实验4组每组6只动物。实验组分别于第1d下午4点,第2d下午4点,第3d上午8点和下午2点手术。第3d下午4点处死全部动物并取材。模型制备:细针穿刺胆总管注射标准致病菌(大肠杆菌ATCC-25922,美国菌种保藏中心提供)025mL,菌液浓度为1×108/mL,不全结扎胆总管(注射菌液后于针上结扎胆总管,再退针)。取材与组化染色:所有动物均于同一部分用锐利刀片切取绿豆大小两块肝组织,制成冰冻切片,以还原型辅酶Ⅱ黄递酶组织化学法对切片进行染色。图像分析:采用MPIAS-500多媒体彩色病理图像分析系统。结果:对照组切片低倍镜(×100)视野所见为正常肝细胞,未见阳性细胞。感染2h组可在中央静脉周围见到环形排列的着浅蓝色的弱阳性细胞及少数着深蓝色的强阳性细胞。感染8h组在中央静脉周围可见大量着深蓝色的强阳性细胞,以中央静脉为中?
