采用NaCl抽提、硫酸铵分部沉淀和 2步离子交换法从黑皮扁豆中纯化得到一种甘露糖专一凝集素 (DPL)。在非变性电泳中纯化的DPL只有一条蛋白带 ,在还原及非还原条件下的SDS PAGE中呈现两条蛋白带 ,其分子量分别为 36 .8kD和 39.8kD ;利用FPLC经SephacrylS 30 0凝胶过滤测得其表观分子量为 15 5kD ,表明DPL由 2个α亚基和 2个 β亚基组成 ,无二硫键。DPL的等电点为 pH 6 .18,含1.6 %糖 ,为酸性糖蛋白。DPL的热稳定性和pH稳定性 ,与已报道的甘露糖 /葡萄糖专一的豆科凝集素相似。
A mannose specific agglutinin (DPL) was purified from black lentils by NaCl extraction, ammonium sulfate fraction precipitation and two-step ion exchange. The purified DPL in non-denaturing electrophoresis had only one protein band and two protein bands in SDS PAGE under reducing and non-reducing conditions, with molecular weights of 36.8kD and 39.8kD, respectively. Sephacryls 30O gel filtration using FPLC Its apparent molecular weight was measured to be 15 5 kD, indicating that DPL consists of two α subunits and two β subunits without disulfide bonds. The isoelectric point of DPL is pH 6.18, containing 1.6% sugar, an acid glycoprotein. The thermal and pH stability of DPL is similar to that reported for mannose / glucose specific lectins.
