采用免疫组化ABC法观察各型大肠癌、大肠绒毛状乳头状瘤和癌切端大肠粘膜组织蛋白酶D的分布、超微结构变化及其与Ⅰ、Ⅱ型胶原关系。结果提示:第Ⅲ型粘液细胞癌(印戒细胞场)组织蛋白酶D分布于胞浆,所形成的粘液池内粘液亦呈阳性,且靠近印戒细胞周围更强。管状腺癌胞核丰富者多为阴性,低分化腺场胞浆丰富者则组织蛋白酶D弥漫于胞浆。粘液池内胶原溶解、消失。电镜下,可见第Ⅲ型粘液细胞场分泌功能旺盛,癌细胞附近亦有间质胶原溶解。提示组织蛋白酶D在大肠癌浸润中能降解间质Ⅰ、Ⅲ型胶原,故粘液细胞癌浸润性强与该酶活性有关。
Immunohistochemical ABC method was used to observe the distribution and ultrastructural changes of cathepsin D in colorectal villous papilloma and large intestine mucosa of the carcinoma, and its relationship with type I and type II collagen. The results suggest that cathepsin D is distributed in the cytoplasm of type III mucinous cell carcinoma (signet-ring cell field), the mucus formed in the mucus pool is also positive, and it is stronger around the signet ring cells. Tubular adenocarcinoma nuclei were mostly negative, while poorly differentiated glandular cells were rich in cathepsin D diffuse in the cytoplasm. The collagen in the mucus pool dissolved and disappeared. Under electron microscope, the secretion of type III mucous cell field was prosperous, and interstitial collagen was also dissolved near the cancer cells. It is suggested that cathepsin D can degrade mesenchymal type I and type III collagen in the invasion of colorectal cancer, so the invasiveness of mucinous cell carcinoma is related to the enzyme activity.
