目的研究甲基乙二醛(MG)对体外培养的海马神经干细胞(NSCs)的影响,以及中药脑康Ⅱ号的干预情况。方法体外培养成年大鼠海马干细胞,分别使用4μmol MG和脑康Ⅱ号含药血清等干预48 h后,进行Nestin、β-Ⅲ-tubulin和DAPI免疫荧光染色,使用Image J软件计算分化的神经元轴突长度。结果Nestin标记的各组细胞的神经轴突没有显著差异;MG组中β-Ⅲ-tubulin阳性染色显著减少,MG组的神经元轴突显著短于其他组(P<0.05);DAPI染色中未发现有核碎裂。结论MG蓄积后有可能通过抑制海马干细胞分化的途径而与糖尿病认知障碍的发病形成关联;而中药脑康Ⅱ号能够有效抑制MG的这种神经毒性,促进海马NSCs的分化。
Objective To investigate the effects of methylglyoxal (MG) on hippocampal neural stem cells (NSCs) cultured in vitro and the intervention of the Chinese herbal medicine Brain Health II. Methods Adult rat hippocampal stem cells were cultured in vitro. After intervention with 4 μmol MG and Naokang II-containing serum for 48 h, immunofluorescence staining of Nestin, β-III-tubulin, and DAPI was performed. Differentiated neurons were calculated using Image J software. Axon length. Results There was no significant difference in the axons of Nestin-labeled cells. The positive staining of β-III-tubulin was significantly decreased in the MG group. The axons of the MG group were significantly shorter than the other groups (P<0.05); Nuclear fragmentation was found. Conclusions MG accumulation may be associated with the pathogenesis of diabetes mellitus by inhibiting the differentiation of hippocampal stem cells. The Chinese herbal medicine, Xiaokang II, can effectively inhibit this neurotoxicity of MG and promote the differentiation of hippocampal NSCs.
