目的:探讨葛根黄酮提取物对HL-60细胞bcl-2和bax基因表达的影响。方法:用RT-PCR和蛋白质免疫印迹技术检测葛根黄酮提取物、葛根素和大豆甙元处理HL-60后,对bcl-2和bax基因的mRNA和蛋白表达的影响。结果:葛根黄酮提取物、葛根素和大豆甙元处理HL-60细胞后能够上调baxmRNA表达水平,下调bcl-2蛋白表达和上调Bax蛋白表达,且随着处理时间的延长其诱导下调和上调能力增强。结论:葛根黄酮提取物诱导HL-60细胞凋亡与其下调bcl-2mRNA和蛋白表达水平和其上调baxmRNA和蛋白表达水平有关。
Objective: To investigate the effect of pueraria flavonoid extract on the expression of bcl-2 and bax genes in HL-60 cells. METHODS: The mRNA and protein expression of bcl-2 and bax genes in HL-60 treated with pueraria flavonoid extract, puerarin and daidzein were detected by RT-PCR and Western blot. RESULTS: After treatment with pueraria flavonoids extract, puerarin and daidzein, HL-60 cells could up-regulate the expression of bax mRNA, down-regulate the expression of bcl-2 protein and up-regulate the expression of Bax protein, and induced down-regulation and up-regulation with the prolongation of treatment time. Enhanced. Conclusion: The apoptosis of HL-60 cells induced by pueraria flavonoids extract is related to the down-regulation of bcl-2 mRNA and protein expression and the up-regulation of bax mRNA and protein expression.
