目的探讨采用RNA干扰(RNA interference,RNAi)技术干扰巨噬细胞集落刺激因子(macrophage colonystimulating factor,M-CSF)抑制子宫内膜异位症(endometriosis,EMs)基质细胞迁移的可行性,并阐明其分子机制。方法异位子宫内膜组织取自深圳市罗湖区人民医院妇科微创治疗中心2014年1月至6月就诊的患者。取组织后分离培养EMs基质细胞,采用小干扰RNA(small interfering RNA,si RNA)敲低M-CSF表达,通过划痕实验检测EMs基质细胞的迁移能力,通过明胶酶谱及实时荧光定量PCR检测基质金属蛋白酶2,9(matrix metalloproteinase,MMP-2,9)的活性及表达。结果敲低M-CSF后显著抑制EMs基质细胞迁移,且抑制该细胞MMP-2及MMP-9的表达及活性。结论采用RNAi技术敲低M-CSF表达很可能是治疗EMs的新方法。
OBJECTIVE: To investigate the feasibility of interfering with the migration of macrophage colony stimulating factor (M-CSF) in endometriosis (EMs) stromal cells by RNA interference (RNAi) Molecular mechanism. Methods Ectopic endometrial tissues were obtained from patients who were treated by gynecological minimally invasive treatment center of People’s Hospital, Luohu District, Shenzhen from January to June 2014. EMs stromal cells were isolated and cultured. The expression of M-CSF was knocked down by small interfering RNA (siRNA), and the migration of EMs stromal cells was detected by scratch test. The gelatin zymography and real-time fluorescence quantitative PCR Activity and Expression of Matrix Metalloproteinase 2 and 9. Results M-CSF knockdown significantly inhibited EMs stromal cell migration and inhibited MMP-2 and MMP-9 expression and activity. Conclusion The knockdown of M-CSF expression by RNAi is likely to be a new method for the treatment of EMs.
