目的:建立血浆中伏立康唑的液相色谱测定方法。方法:采用甲醇沉淀处理血浆样品,采用高效液相色谱紫外检测法测定。色谱柱为Diamonsil C18(4.6mm×250mm,5μm),流动相为水-乙腈-冰醋酸-三乙胺(60∶40∶0.1∶0.05),紫外波长258nm,流速1.0mL·min-1,柱温25℃。结果:标准曲线的线性范围为0.055~4.4mg·L-1,r=0.9999(n=5),低、中、高3种浓度的相对回收率分别为97.3%,100.5%,99.4%,日内和日间RSD均小于10%。结论:本方法简便,快速,准确,专一性好,可用于伏立康唑的血药浓度测定和药动学研究。
Objective: To establish a method for the determination of voriconazole in plasma by liquid chromatography. Methods: The plasma samples were treated with methanol precipitation and determined by high performance liquid chromatography with UV detection. The column was Diamonsil C18 (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of water-acetonitrile-glacial acetic acid-triethylamine (60:40:0.1:0.05), UV wavelength was 258 nm and flow rate was 1.0 mL · min- Temperature 25 ℃. Results: The linear range of the standard curve was 0.055 ~ 4.4 mg · L-1, r = 0.9999 (n = 5). The relative recoveries of the three concentrations were 97.3%, 100.5% and 99.4% And daytime RSDs were less than 10%. Conclusion: The method is simple, rapid, accurate and specific and can be used for the determination of voriconazole and pharmacokinetics.
