目的测定磷酸川芎嗪的血浆蛋白结合率。方法采用微透析法测定磷酸川芎嗪的血浆蛋白结合率,用反透析法进行探针回收率校正。血浆蛋白结合率用f=(ρ0-ρf)/ρ0计算。结果磷酸川芎嗪与牛血清白蛋白、人血清白蛋白和人血浆、大鼠血浆的蛋白结合率分别为(79.51±1.66)%、(68.34±1.43)%、(56.17±1.44)%和(70.97±2.19)%。结论磷酸川芎嗪与血浆蛋白具有中等强度的结合,采用微透析进行血浆蛋白结合率研究可行。
Objective To determine the plasma protein binding rate of ligustrazine phosphate. Methods The plasma protein binding rate of ligustrazine phosphate was determined by microdialysis method and the probe recovery was corrected by reverse dialysis method. Plasma protein binding rate with f = (ρ0-ρf) / ρ0 calculation. Results The protein binding rates of ligustrazine phosphate and bovine serum albumin, human serum albumin, human plasma and rat plasma were (79.51 ± 1.66)%, (68.34 ± 1.43)%, (56.17 ± 1.44)% and (70.97 ± 2.19)%. Conclusion Tetramethylpyrazine phosphate has a moderate binding to plasma protein. It is feasible to study the plasma protein binding rate using microdialysis.
