目的探讨坏死性小肠结肠炎(NEC)诱导的全身炎症反应综合征(SIRS)动物模型血浆细胞因子水平。方法 40只新生Wistar大鼠,日龄36~48 h,随机分为对照组10只及NEC模型组30只。模型组大鼠采用低温缺氧、脂多糖10 mg/kg灌胃制备NEC模型,鼠乳代用品人工喂养,对照组采用生理盐水灌胃,不进行低温和缺氧。实验结束后处死动物,模型组根据血液白细胞总数及呼吸改变确定是否发生SIRS,并进一步分为未发生SIRS组和发生SIRS组。采用液相芯片技术比较血浆白细胞介素(IL)-1β、肿瘤坏死因子(TNF)-α、IL-6和IL-10的含量。结果与对照组比较,模型组血浆细胞因子IL-1β、TNF-α、IL-6、IL-10水平均升高,且发生SIRS组细胞因子水平高于未发生SIRS组(P均<0.05)。对照组、未发生SIRS组和发生SIRS组各细胞因子水平分别为:IL-1β(0.100±0.016)、(3.187±0.525)、(4.182±0.552)pg/ml;TNF-α(0.027±0.008)、(0.854±0.101)、(1.077±0.133)pg/ml;IL-6(0.033±0.006)、(0.984±0.102)、(2.039±0.483)pg/ml;IL-10(0.056±0.015)、(1.978±0.096)、(2.810±0.435)pg/ml。结论 NEC诱导的SIRS新生大鼠模型中,血浆IL-1β、TNF-α、IL-6、IL-10含量升高,提示细胞因子之间的相互调节可能影响SIRS的进程。
Objective To investigate the levels of plasma cytokines in animal models of systemic inflammatory response syndrome (SIRS) induced by necrotizing enterocolitis (NEC). Methods Forty neonatal Wistar rats were randomly divided into control group (n = 10) and NEC model group (n = 30). The rats in the model group were given NAC model by intragastric administration of hypoxia and lipopolysaccharide 10 mg / kg. Rats in the control group were fed with saline instead of hypothermia and hypoxia. The animals were sacrificed after the experiment, the model group according to the total number of leukocytes and respiratory changes to determine whether the occurrence of SIRS, and further divided into no SIRS group and the occurrence of SIRS group. The levels of plasma interleukin (IL) -1β, tumor necrosis factor (TNF) -α, IL-6 and IL-10 were compared by liquid chromatography. Results Compared with the control group, the levels of cytokines IL-1β, TNF-α, IL-6 and IL-10 in the model group were increased, and the levels of cytokines in the SIRS group were higher than those without the SIRS group (all P <0.05) . The levels of IL-1β (0.100 ± 0.016), (3.187 ± 0.525), (4.182 ± 0.552) pg / ml and TNF-α (0.027 ± 0.008) in control group, SIRS group and SIRS group were , (0.854 ± 0.101), (1.077 ± 0.133) pg / ml, IL-6 (0.033 ± 0.006), (0.984 ± 0.102), (2.039 ± 0.483) pg / 1.978 ± 0.096), (2.810 ± 0.435) pg / ml. Conclusion The levels of IL-1β, TNF-α, IL-6 and IL-10 in the NEC-induced neonatal rat model of SIRS are increased, indicating that the mutual regulation of cytokines may influence the course of SIRS.
