目的观察大鼠颈总动脉球囊损伤后血凝素样氧化低密度脂蛋白受体-1(lectin like oxidized low density lipoprotein receptor-1,LOX-1)的表达及西洛他唑对其表达的影响,探索西洛他唑抑制内膜增生的机制。方法24只健康雄性SD大鼠按完全随机分组法分成假手术组、模型组、西洛他唑组(n=8)。观察球囊损伤后内膜增生情况并计算内膜、中膜面积比值,检测血清MDA含量,RT-PCR法和免疫组化法分别检测各组球囊损伤后大鼠颈总动脉血管壁组织中LOX-1mRNA和蛋白的表达。结果西洛他唑组内膜/中膜面积比值(IA/MA)、LOX-1mRNA及蛋白表达水平、血清MDA含量显著低于模型组,差异有统计学意义(P<0.05)。结论西洛他唑可显著抑制内膜增生,抑制LOX-1表达及其介导的氧化应激,这可能是西洛他唑抑制内膜增生的机制之一。
Objective To observe the expression of lectin like oxidized low density lipoprotein receptor-1 (LOX-1) after balloon injury of common carotid artery in rats and the effect of cilostazol on its expression Influence, explore cilostazol inhibition of intimal hyperplasia mechanism. Methods Twenty-four healthy male Sprague-Dawley rats were randomly divided into sham operation group, model group and cilostazol group (n = 8). The intimal hyperplasia was observed after balloon injury and the ratio of intima to media was calculated. The content of serum MDA was measured. The contents of MDA in serum were detected by RT-PCR and immunohistochemistry. LOX-1 mRNA and protein expression. Results The ratio of intima / media area (IA / MA), LOX-1 mRNA and protein, and serum MDA in cilostazol group were significantly lower than those in model group (P <0.05). Conclusion Cilostazol can significantly inhibit intimal hyperplasia and inhibit the expression of LOX-1 and its oxidative stress, which may be one of the mechanisms of cilostazol in inhibiting intimal hyperplasia.
