目的:建立白花蛇舌草中齐墩果酸和熊果酸的含量测定方法。方法:采用毛细管区带电泳,高频电导检测;未涂层弹性融硅石英毛细管柱55 cm×75μm ID,有效长度50 cm;1.2 mmol/L三乙胺-HCl(pH=10.0),0.24mmol/Lβ-环糊精为运行缓冲液;分离电压12.5 kV;重力进样10 s(高度20 cm)。结果:以布洛芬为内标,齐墩果酸、熊果酸线性范围分别为3.9~39.0μg/ml(r=0.9991)和20.0~140.0μg/ml(r=0.9990);平均回收率分别为97.1%和96.0%。结论:该法简便、准确、快速、重现性好,可用于白花蛇舌草药材中齐墩果酸和熊果酸的含量测定。
Objective: To establish a method for determination of oleanolic acid and ursolic acid in Hedyotis diffusa. Methods: Capillary zone electrophoresis, high frequency conductivity detection; uncoated elastic silica-silica capillary column 55 cm×75 μm ID, effective length 50 cm; 1.2 mmol/L triethylamine-HCl (pH=10.0), 0.24 mmol The /Lβ-cyclodextrin was used as the running buffer; the separation voltage was 12.5 kV; the gravity injection was 10 s (height 20 cm). Results: With ibuprofen as internal standard, the linear ranges of oleanolic acid and ursolic acid were 3.9~39.0μg/ml (r=0.9991) and 20.0~140.0μg/ml (r=0.9990) respectively; the average recoveries were respectively 97.1% and 96.0%. Conclusion: The method is simple, accurate, rapid, and reproducible. It can be used to determine the content of oleanolic acid and ursolic acid in snake tongue.
