以胸腺肽α_1标准品为对照品,用高效毛细管电泳法测定胸腺肽浴液中α_1的含量。使用未涂层石英毛细管柱(78cm×30μm);电泳缓冲溶液为0.05mol/L磷酸盐缓冲液(pH7.0);运行电压15KV;负压进样(66.7KPa);温度25℃;检测波长200nm;进样时间1秒。α_1标准品在浓度50~800μg/ml范围内呈线性关系,回归方程Y=1.76×10~3×3.54,r=0.9998低中高三种浓度(50,200,800μg/ml)的回收率分别为87%,91%和95%。用比例色谱峰的方法确定胸腺肽溶液中α_1峰的纯度,测得三批胸腺肽溶液中α_1的质量百分含量分别为1.24%,0.812%和0.732%。
Thymosin α_1 standard was used as a reference substance, and the content of α_1 in thymosin bath was determined by high performance capillary electrophoresis. The purity of α_1 peak in thymosin solution was determined by proportional chromatographic peak method. The mass percent of α_1 in three batches of thymosin solution was 1.24%, 0.812% and 0.732%, respectively.
