目的由线栓法大脑中动脉闭塞致脑缺血叠加寒冷诱导建立血瘀证表征模型,通过与寒凝血瘀证表征模型和脑缺血诱导血瘀证表征模型比较,选择一种最合适的血瘀证表征模型。方法选择大脑中动脉闭塞(MCAO)大鼠模型、冰水冷冻大鼠模型、大脑中动脉闭塞叠冰水冷冻大鼠模型,进行脑缺血后1、3、5、7d不同时间点大鼠证候模型表征变化(舌、舌下脉络)及其对模型大鼠进行行为学、血液流变学检测。结果脑缺血后3dMCAO组、冰水组与冰水MCAO组大鼠的舌质色度值与正常组比较均存在统计学差异(P<0.05);脑缺血后5d冰水MCAO组大鼠的舌质色度值与正常组比较存在统计学差异(P<0.05);脑缺血后3d冰水MCAO组舌质色度值为各时间点最高值(舌质色度值60.12±5.15),但各组间舌质色度值比较统计学无差异(P>0.05)。各组血液流变学统计结果显示,MCAO组、冰水冷冻组、冰水MCAO组全血及还原黏度、血球压积、血浆黏度、聚集指数、变形指数均较正常组升高,部分时间点值升高与正常组比较有统计学差异(P<0.01,P<0.05),其中又以冰水MCAO组脑缺血后3d血液黏度、血球压积、血浆黏度、聚集指数均为各组最高值,变形指数为各组最低值。结论在3种模型中,大脑中动脉闭塞叠加寒冷模型可能为最合适的诱导血瘀证表征模型的动物模型。
Objective To establish a model of blood stasis syndrome induced by occlusion of middle cerebral artery occlusion caused by occlusion of the middle cerebral artery occlusion and compare with the characterization model of cold stasis blood stasis syndrome and blood stasis syndrome induced by cerebral ischemia to select the most suitable blood Stasis syndrome characterization model. Methods The rat models of middle cerebral artery occlusion (MCAO), ice-water freezing rat model and middle cerebral artery occlusion ice-water freezing rat model were selected. Rats in different time points at 1, 3, 5 and 7 days after cerebral ischemia Model changes in the waiting list (tongue, sublingual vein) and its behavior in rats model, hemorheology test. Results Compared with the normal group, the tongue colorimetric values of MCAO group, ice-water group and icy water MCAO group were significantly different after cerebral ischemia (P <0.05) (P <0.05). The values of tongue color of MCAO group were the highest (60.12 ± 5.15) at each time point after cerebral ischemia , But no significant difference was found in the tongue colorimetry between the groups (P> 0.05). The results of hemorheology in each group showed that whole blood and reducing viscosity, hematocrit, plasma viscosity, aggregation index and deformation index in MCAO group, ice-water freezing group and ice-cold MCAO group were all higher than those in normal group, and part of time points (P <0.01, P <0.05). The blood viscosity, hematocrit, plasma viscosity and aggregation index of MCAO group were the highest Value, deformation index for each group the lowest value. Conclusions In the three models, the middle cerebral artery occlusion superimposed cold model may be the most suitable animal model for inducing blood stasis syndrome characterization model.
