目的 研究热休克蛋白 70在皮肤癌中的表达水平和分布特点及与增殖细胞核抗原 (PCNA)、 p53蛋白表达的相关性。方法 应用免疫组化方法 (SABC),用特异性热休克蛋白 70单抗、 p53蛋白单抗、 PCNA单抗检测热休克蛋白 70、 PCNA、 p53蛋白在皮肤良恶性病变和正常皮肤组织中的表达和分布。 结果 热休克蛋白 70在鳞状细胞癌、基底细胞癌癌细胞核内有异常增高表达,其中鳞状细胞癌、基底细胞癌中阳性与阴性标本的 PC10指数差异有显著性,而 p53蛋白阳性率未见明显差异。鳞状细胞癌中热休克蛋白 70表达在Ⅰ~Ⅱ 和Ⅳ级间差异有显著性。结论 热休克蛋白 70在皮肤癌中存在异常增高的核表达,并与细胞的增殖程度和病理分级有关,提示其可能参与肿瘤生长,其在皮肤肿瘤中的诊断价值和与临床预后的关系需要进一步研究。
Objective To study the expression and distribution characteristics of heat shock protein 70 in skin cancer and its correlation with PCNA and p53 protein expression. Methods Immunohistochemical method (SABC) was used to detect the expression of heat shock protein 70, PCNA and p53 protein in benign and malignant skin lesions and normal skin tissues with specific heat shock protein 70 monoclonal antibody, p53 monoclonal antibody and PCNA monoclonal antibody. And distribution. Results The expression of heat shock protein 70 was abnormally increased in squamous cell carcinoma and basal cell carcinoma cell nucleus. The PC10 index of positive and negative specimens in squamous cell carcinoma and basal cell carcinoma were significantly different, but the positive rate of p53 protein was not. See the obvious difference. The expression of heat shock protein 70 in squamous cell carcinoma was significantly different between grades I-II and IV. Conclusion Heat shock protein 70 has abnormally increased nuclear expression in skin cancer, which is associated with cell proliferation and pathological grade, suggesting that it may be involved in tumor growth. Its diagnostic value in skin tumors and its relationship with clinical prognosis need further the study.
