在长期的病理制片工作中,常常会出现组织脱水不彻底、脱片和缺刀等问题,我们通过反复研究,总结出几条小经验,现介绍如下:1 微波梯度酒精脱水传统的脱水方法每缸需1小时,冬季气温低或组织较致密时(如子宫),脱水时固定剂不易渗透到组织内面,既使勤换固定剂也无效,此时必须延长脱水时间,这种方法导致:(1)费时,制片周期延长;(2)固定剂放置时间过长,使某些特异性抗原变性、丢失,做免疫组化染色时,出现假阴性.我们将常规酒精梯度脱水改为微波梯度酒精脱水,利用微波辐射提高组织细胞通透性和溶剂分子穿透能力,达到组织处理的目的.其方法为:把组织块放入酒精缸内,然后把酒精缸放入微波内,用微波4档(功率 300W)依次幅射,每缸5min(微波幅射时酒精缸应盖盖).我们体会到这种方法的优点:(1)脱水彻底;(2)组织处理时间缩短,制片周期也相应缩短;(3)减少了抗原的丢失和损害,对做免疫组化染色有较好的效
