目的探讨口服西地那非对老龄大鼠心脏及阴茎组织凋亡的影响。方法 30只大鼠平均分为两组,实验组给予西地那非灌胃(12 mg/kg),对照组生理盐水灌胃(1.6 ml/kg),每周3次,共8周。取心脏及阴茎组织,检测诱导型一氧化氮合酶(induced nitricoxide synthase,iNOS)的表达及细胞凋亡率。结果实验组灌胃后抓腮、爬高、追逐动作明显增多。实验组、对照组心脏组织iNOS表达的平均吸光度(optical density,OD)值分别为(0.32±0.04)、(0.40±0.05)(P>0.05),细胞凋亡率分别为(3.65±1.20)%、(2.81±0.78)%(P>0.05);阴茎组织iNOS的OD值分别为(0.37±0.04)、(0.23±0.03)(P<0.05),细胞凋亡率分别为(3.95±0.72)%、(1.43±0.69)%(P<0.01)。结论老龄大鼠长期服用西地那非后性行为增多,心脏组织iNOS表达及细胞凋亡变化不大;阴茎组织iNOS表达增多,细胞凋亡现象明显增加。
Objective To investigate the effect of oral sildenafil on the apoptosis of the heart and penile tissue in aged rats. Methods Thirty rats were equally divided into two groups. The experimental group was given sildenafil (12 mg / kg) and the control group was given normal saline (1.6 ml / kg) three times a week for 8 weeks. The heart and penile tissues were taken for detecting the expression of induced nitric oxide synthase (iNOS) and the rate of apoptosis. Results The experimental group after gavage grasping, climbing high, the chase movements increased significantly. The average optical density (OD) values of iNOS in experimental and control groups were (0.32 ± 0.04) and (0.40 ± 0.05), respectively (P> 0.05), and the apoptotic rates were (3.65 ± 1.20)% , (2.81 ± 0.78)% respectively (P> 0.05). The OD of iNOS in penile tissue were (0.37 ± 0.04) and (0.23 ± 0.03) , (1.43 ± 0.69)% (P <0.01). Conclusions After long-term use of sildenafil in aged rats, the sexual behavior is increased, the expression of iNOS and the apoptosis in heart tissue are not changed much. The expression of iNOS in penile tissue is increased and the apoptosis of cells is obviously increased.
