从市售阿城紫皮蒜生长的叶片经PEC沉淀差速离心提纯,获得复合病毒制剂,得率为4.90mm(100g)-1叶。电镜观察病毒粒体为长度550~800nm的线状物,经抗血清测定主要为洋葱黄矮病毒(OYDV)和大蒜潜隐病毒(GLV)。用复合病毒制剂免疫制备的抗血清效价为1024~2048。OYDV抗血清检测茎尖脱毒组培苗,脱毒率为50%左右,用GLV抗血清检测,脱毒率为95%~97%,复合病毒抗血清检测结果与OYDV相同。证明复合病毒抗血清可用于组培苗的检测。组培脱毒大蒜在田间种植1年后发病率为50%,3年后达80%。茎尖组培脱毒率不高。需用抗血清逐株检测挑选,淘汰病苗,才能获得健康无毒的大蒜群体。
The leaves from the commercially available Acheng purple garlic were purified by PEC precipitation differential centrifugation to obtain a composite virus preparation with a yield of 4.90 mm (100 g) -1 leaves. Electron microscopy showed that the virus particles were linear in length of 550-800 nm. The anti-serum was mainly OYDV and GLV. The titer of the antiserum prepared with the compound virus preparation was 1024-2048. OYDV antiserum detected tips tip detoxification tissue culture, detoxification rate of about 50%, with GLV antiserum detection, detoxification rate of 95% to 97%, the composite virus antiserum test results and OYDV the same. Proved that the composite virus antiserum can be used for tissue culture test. Tissue culture virus-free garlic in the field one year after the incidence of 50%, up to 80% after 3 years. Shoot tip tissue culture off rate is not high. Need to use antisera testing and selection of each strain, out of disease vaccine, in order to obtain healthy and non-toxic groups of garlic.
