目的:建立毛细管区带电泳内标法测定半夏鸟苷含量的方法。方法:采用毛细管区带电泳内标法定量,以熔融石英毛细管(57 cm×75μm,有效长度50 cm)为分离通道,运行缓冲液经NaOH溶液调pH至9.0的0.8 mol.L-1硼酸溶液-0.1mol.L-1硼砂溶液-乙腈(50∶50∶13),分离电压25 kV,检测波长254 nm,毛细管温度25℃,自动压力进样5 s。结果:半夏中鸟苷在0.25~1.5 mg线性关系良好(r=0.999 1),鸟苷加样回收率为102.0%,RSD为2.4%。结论:用毛细管区带电泳内标法测定半夏鸟苷含量的方法准确可行。
Objective: To establish a method for the determination of the content of Pinellin by capillary zone electrophoresis with internal standard method. Methods: The capillary zone electrophoresis was used to quantify the internal standard. The fused silica capillary tube (57 cm × 75 μm, effective length 50 cm) was used as the separation channel. The buffer was adjusted to pH 8.0 with 0.8 mol·L-1 boric acid solution -0.1mol.L-1 borax solution - acetonitrile (50:50:13), separation voltage 25 kV, detection wavelength 254 nm, capillary temperature 25 ℃, automatic pressure injection 5 s. Results: Guanosine had a good linearity (r = 0.999 1) between 0.25 and 1.5 mg in perennial. The recovery of guanosine was 102.0% and the RSD was 2.4%. Conclusion: The method of capillary zone electrophoresis with internal standard method for the determination of Pinellia glycosides content is accurate and feasible.
