酸枣仁汤对老年慢性睡眠剥夺大鼠视交叉上核中血管活性肠肽、精氨酸加压素肽能神经细胞的影响

目的:研究酸枣仁汤对老年慢性睡眠剥夺大鼠视交叉上核中血管活性肠肽(VIP)、精氨酸加压素(AVP)肽能神经细胞的影响。方法:利用“多平台水环境法”建立老年大鼠慢性睡眠剥夺模型,将实验大鼠随机分为4组,环境对照组,慢性睡眠剥夺模型组,舒乐安定组以及酸枣仁汤组。用免疫组织化学方法观察睡眠剥夺21 d后各组大鼠视交叉上核中VIP、AVP肽能神经细胞的变化,用Real Time-PCR法检测视交叉上核中VIP mRNA、AVP mRNA的表达。结果:与环境对照组相比较,慢性睡眠剥夺模型组VIP表达明显降低、AVP表达明显升高(P<0.05);与慢性睡眠剥夺模型组比较,治疗组VIP则有不同程度的升高(P<0.05),AVP则出现明显下降,其中酸枣仁汤组上下浮动最为显著。与环境对照组比较,慢性睡眠剥夺模型组VIP、AVPmRNA表达下调;与慢性睡眠剥夺模型组比较,舒乐安定组和酸枣仁汤组VIP、AVPmRNA表达上调。结论:酸枣仁汤能够调控大脑视交叉上核VIP、AVP的表达,有效调节老年睡眠剥夺大鼠的昼夜节律。 Objective: To study the effects of Suanzaoren Decoction on vasoactive intestinal peptide (VIP) and arginine vasopressin (AVP) peptide neurons in the suprachiasmatic nucleus of aged rats with chronic sleep deprivation. Methods: The model of chronic sleep deprivation in aged rats was established by using “multi-platform water environment method”. The rats were randomly divided into 4 groups: environmental control group, chronic sleep deprivation model group, lelerazepam group, . Immunohistochemistry was used to observe the changes of VIP and AVP peptide neurons in the suprachiasmatic nucleus of rats 21 days after sleep deprivation. The expression of VIP mRNA and AVP mRNA in the suprachiasmatic nucleus was detected by Real Time-PCR. Results: Compared with the environmental control group, the expression of VIP in chronic sleep deprivation model group was significantly decreased and the expression of AVP was significantly increased (P <0.05). Compared with chronic sleep deprivation model group, the VIP in treatment group was increased to some extent <0.05), AVP was significantly decreased, of which the Suanzaoren soup group floating up and down the most significant. Compared with the environmental control group, the expression of VIP and AVP mRNA in chronic sleep deprivation model group was down-regulated. Compared with chronic sleep deprivation model group, VIP and AVP mRNA expression in Shule An Anding group and Suanzaoren Decoction group were up-regulated. Conclusion: Suanzaoren decoction can regulate the expression of VIP and AVP in the suprachiasmatic nucleus of the brain and regulate the circadian rhythm of aged rats with sleep deprivation.
