目的 :了解粒细胞集落刺激因子 (G CSF)对人卵巢癌细胞株生长的影响 ,并对用于卵巢癌细胞活性检测的 4种方法进行比较。方法 :采用XTT比色法、MTT比色法、集落形成试验、3H TdR掺入试验分别检测G CSF对卵巢癌细胞株 3AO细胞生长的影响。结果 :4种细胞活性检测方法检测不同浓度G CSF对 3AO细胞生长的影响与对照组 (G CSF浓度为 0ng/ml)相比 ,差异均无显著性 (P >0 0 5 )。结论 :G CSF对 3AO细胞生长无显著影响 ,对其可促进卵巢癌细胞增殖的顾虑也许是不必要的 ;XTT比色法可替代其他方法用于卵巢癌细胞活性检测 ,且敏感性更高、更加快速简便。
Objective: To investigate the effect of granulocyte colony-stimulating factor (G CSF) on the growth of human ovarian cancer cell lines and to compare the four methods used to detect ovarian cancer cell activity. Methods: The effects of G CSF on the growth of ovarian cancer cell line 3AO were detected by XTT assay, MTT assay, colony formation assay and 3H TdR incorporation assay. Results: There was no significant difference between the four cell activity test methods in detecting the effect of different concentrations of G CSF on the growth of 3AO cells compared with that of the control group (Gng concentration 0 ng / ml) (P> 0.05). CONCLUSIONS: G CSF has no significant effect on the growth of 3AO cells, and it may be unnecessary for G CSF to promote the proliferation of ovarian cancer cells. XTT can replace other methods for the detection of ovarian cancer cell activity and is more sensitive, More quick and easy.
