本研究通过化学诱癌剂DMEA诱发金黄地鼠颊囊粘膜鳞癌,建立动物模型,经鱼精蛋白及联合平阳霉素治疗,采用组织学定量方法,同时检测肿瘤内血管密度与血管面积密度。结果发现鱼精蛋白具有明显抑制地鼠颊囊癌生长的作用,其治疗效果来源于对肿瘤血管生成的抑制,肿瘤生长早期使用鱼精蛋白进行治疗,具有明显的疗效。平阳霉素的抑瘤作用来自其对肿瘤细胞的杀伤,它对肿瘤血管生成无明显抑制作用。联合应用抑制血管生成及直接细胞毒作用的两种不同作用机制的抗癌药物治疗地鼠颊囊癌有显著协同疗效,可望成为临床上治疗颊癌的有效手段之一。
In this study, golden hamster cheek pouch squamous cell carcinoma was induced by chemoattractant agent DMEA. Animal model was established by protamine and pingyangmycin. Histological and quantitative methods were used to detect the intracranial vascular density and vascular density. The results showed that protamine has a significant inhibitory effect on the growth of hamster cheek pouch cancer, the therapeutic effect derived from the inhibition of tumor angiogenesis, the use of protamine for the treatment of early tumor growth has obvious curative effect. The anti-tumor effect of Pingyangmycin comes from its killing of tumor cells, and it has no obvious inhibitory effect on tumor angiogenesis. Combined application of two different mechanisms of anti-angiogenesis and direct cytotoxicity of anti-cancer drugs in treating hamster cheek pouch have a significant synergistic effect, is expected to become clinically effective treatment of buccal cancer.
