目的探讨仙灵脾改善2型糖尿病(T2DM)大鼠胰岛素抵抗的机制。方法 70只Wistar大鼠,随机选取10只作为正常对照组,予普通饲料喂养;60只为造模组,采用高脂、高糖饲料加小剂量链脲佐菌素建立T2DM大鼠模型,其中成模的44只大鼠随机分为T2DM模型组16只、仙灵脾组16只、二甲双胍组12只。仙灵脾组灌服中药仙灵脾3.0g·kg-1·d-1;模型组和正常对照组予等量生理盐水;二甲双胍组予二甲双胍混悬液10mg·kg-1·d-1灌胃给药。用药8周末,取血检测空腹血糖(FBG)、空腹胰岛素(FINS)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、单核细胞趋化蛋白-1(MCP-1),计算胰岛素敏感指数(ISI)。结果与T2DM模型组相比,仙灵脾组、二甲双胍组FBG、FINS、TNF-α、IL-6及MCP-1水平明显降低(P均<0.05),ISI水平明显升高(P均<0.05);与二甲双胍组比较,仙灵脾组FBG较高,ISI较低(P均<0.05)。结论仙灵脾可以通过改善炎症反应来改善T2DM大鼠的胰岛素抵抗。
Objective To investigate the mechanism of Xianling spleen improving insulin resistance in type 2 diabetes mellitus (T2DM) rats. Methods Totally 70 Wistar rats were randomly divided into normal control group and normal control group. Sixty mice were used as model group. T2DM rats were induced by high-fat and high-sugar diet plus low-dose streptozotocin Forty-four rats were randomly divided into T2DM model group, 16 cents spleen group and 12 metformin group. The model group and the normal control group were given the same amount of normal saline; the metformin group was given metformin suspension 10mg · kg-1 · d-1 Stomach administration. At the end of the 8th week after treatment, blood samples were taken for fasting blood test for FBG, FINS, TNF-α, IL-6 and MCP- MCP-1), insulin sensitivity index (ISI) was calculated. Results The levels of FBG, FINS, TNF-α, IL-6 and MCP-1 in Xianling spleen group and metformin group were significantly lower than those in T2DM group (all P <0.05) ); Compared with metformin group, Epimedium group had higher FBG and lower ISI (all P <0.05). Conclusion Epimedium can improve insulin resistance in T2DM rats by improving the inflammatory response.
